- Incorporates into the liposome bilayer, therefore liposome remains fluorescent until bilayer structure is completely dissembled even when liposome contents are released
- Does not readily exchange out of liposomes into cell membranes or other lipid-containing structures (HDL, etc.)
- Detectable by, and functions in, all fluorescence methods (fluoroscopy, fluorescence resonance energy transfer (FRET), etc.) using a rhodamine filter
- Minimal fluorescence in aqueous phase, but high yield when incorporated into lipid bilayers
- DiI liposomes are highly visible to the naked eye even in small quantities
