» Prepared identically to Clodrosome except no clodronate is added and fluorophore is incorporated
» Serves as Clodrosome control because phagocytes can respond immunologically to liposome uptake
» Extruded through 2 µm membranes to ensure that liposomes easily pass through lung capillaries
» Prepared and packaged under sterile conditions
» Some DiI (pronounced “dye-eye”) advantages…

  • Incorporates into the liposome bilayer, therefore liposome remains fluorescent until bilayer structure is completely dissembled even when liposome contents are released
  • Does not readily exchange out of liposomes into cell membranes or other lipid-containing structures (HDL, etc.)
  • Detectable by, and functions in, all fluorescence methods (fluoroscopy, fluorescence resonance energy transfer (FRET), etc.) using a rhodamine filter
  • Minimal  fluorescence in aqueous phase, but high yield when incorporated into lipid bilayers
  • DiI liposomes are highly visible to the naked eye even in small quantities
