L-α-Phosphatidylcholine and Cholesterol

Palmitoyloleoylphosphatidylcholine (POPC) is the predominant naturally-occuring phospholipid and therefore the predominant phospholipid species in egg-derived phosphatidylcholine (>50-60%). The cis double bond in the oleic acid chain of POPC allows rotational motion among the acyl chains of the phospholipid bilayer resulting in the liquid crystalline or fluid phase. Biological membranes take advantage of the fluid phase to continuously reorganize their components into various microdomains allowing the cell membrane regulate the uptake and release of compounds along with the multitude of other functions modulated by cellular membrane components.

Among its many biological functions, cholesterol allows the acyl chains of phospholipids to pack closer together, thus regulating the permeability of lipid bilayers. 30 mole % or greater is the generally accepted concentration of cholesterol which renders fluid-phase liposomes maximally resistant to the disruptive effect of serum proteins on liposomes.
